Мультиформная глиобластома (GBM) является наиболее распространенной и агрессивной формой первичной злокачественной опухоли головного мозга у взрослых с плохим прогнозом. Было показано, что экзосомы являются полезными неинвазивными биомаркерами для диагностики и прогноза опухолей, включая GBM. Экзосомы играют роль в качестве биологических носителей, которые могут выполнять различные задачи через различные сигнальные пути канцерогенеза, такие как PI3K/AKT, SOX2, PTEN, ERK и STAT3.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Экзосомы были выделены из плазмы крови, отобранной у пациентов с диагнозом GBM до хирургической резекции.</p></sec><sec><title>Результаты и обсуждение</title><p>Результаты и обсуждение. Экзосомы, полученные из плазмы крови больных с GBM, имели размеры 40–100 нм и сферическую форму, что соответствует морфологическим характеристикам экзосом. Сочетание ультрафильтрации и двойного ультрацентрифугирования позволяет получить образцы экзосом из плазмы крови без примесей частиц более 100 нм, а форма и размер этих везикул соответствуют характеристикам экзосом, выделенных из других биологических жидкостей.</p></sec><sec><title>Заключение</title><p>Заключение. Описанный здесь экспериментальный протокол для экстракции экзосом из плазмы крови пациентов с GBM является эффективным методом обеспечения чистоты экзосом. Использование этого метода дает возможности для будущих исследований относительно роли экзосом в патогенезе GBM, и его можно было бы в равной степени использовать для исследований c другими патологиями человека.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. Glioblastoma multiforme (GBM) is the most common and aggressive form of primary malignant brain tumour in adults associated with a poor prognosis. Exosomes have been shown to be useful non-invasive biomarkers for the diagnosis and prognosis of tumours, GBM included. Exosomes play a role of biological carriers which can perform various tasks through various signalling pathways of carcinogenesis, such as PI3K/AKT, SOX2, PTEN, ERK and STAT3.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. Exosomes were isolated from blood plasma taken from patients diagnosed with GBM prior to surgical resection.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. Plasma exosomes from patients with GBM had spherical shape and varied in size from 40 to 100 nm matching the exosomes’ morphological characteristics. The combination of ultrafiltration and double ultracentrifugation makes it possible to extract exosome examples from plasma without the presence of contaminating particles over 100 nm in size; the shape and size of these vesicles match the characteristics of exosomes isolated from other biological fluids.</p></sec><sec><title>Conclusion</title><p>Conclusion. The experimental protocol for the extraction of exosomes from GBM patients’ plasma described here proves effective as a method used to ensure the purity of exosomes. Applying this method offers further opportunities for research into the role of exosomes in GBM pathogenesis.
